چکیده
Abstract Novel treatment approaches for pediatric central nervous system (CNS) tumors are urgently needed. A lack of patient-derived tumor cells impedes progress towards developing such new therapies. We intended to overcome this challenge by establishing methods create a biorepository viable single cell suspensions brain surgical specimens. Quantitative and qualitative comparisons tissue processing strategies were performed preserve viability heterogeneous immune cells. drug targets identified analyzing pathways affected RNA transcripts that highly expressed (outliers) in patients’ tumor; outliers each patient determined RNA-seq data from individual compared with compendium 12,747 adult samples harmonized the Treehouse Childhood Cancer Initiative at UCSC Genomics Institute. The predicted anti-tumor efficacy small molecule inhibitors outlier was tested assays against short-term cultured matched patients. Successful collection required obtaining informed consent, standard operating procedures, sample recording using Laboratory Inventory Management Software (LIMS). Since 2020 we have banked 67 CNS specimens Stanford. Amongst those, 51 cases yielded sufficient cryoprotection. most common histology low-grade glioma (LGG, 26 67), majority which pilocytic astrocytoma (18 26). second third types embryonal (6 medulloblastoma, 3 AT/RT) ependymoma (4), respectively. significant differences different preservation media. An pathway previously not implicated LGG sensitivity inhibitor demonstrated. Taken together, established feasibility validating therapeutic vulnerabilities genome-guided approach cultures This works facilitates rapid development personalized treatment.

Alternative specimens for workplace drug testing.
Recent advances in analytical techniques have enabled the detection of drugs and drug metabolites in alternative biological specimens for the purposes of workplace testing. A wide variety of specimens are available, each providing valuable information concerning prior or current drug use. The present focus is on oral fluid (saliva), hair, and sweat. An extensive evaluation by the Division of Wo...
متن کاملMethod for Novel Anti-Cancer Drug Development using Tumor Explants of Surgical Specimens
The current therapies for malignant glioma have only palliative effect. For therapeutic development, one hurdle is the discrepancy of efficacy determined by current drug efficacy tests and the efficacy on patients. Thus, novel and reliable methods for evaluating drug efficacy are warranted in pre-clinical phase. In vitro culture of tumor tissues, including cell lines, has substantial phenotypic...
متن کاملCT-guided laser probe for ablation of brain tumors
In this study, 22 patients (15-75 years old) were selected and transferred to CT scan for tumor ablation. For ablations, after prep and drep under the local anesthesia and mild sedation in proper position, small incision made and special needle inserted and guided by proper direction to the core of the tumor. Then, laser probe inserted through the needle and laser energy delivered. Although we ...
متن کاملImage-Guided Brain Tumor Resection
Surgical treatment of primary brain tumors remains a considerable challenge for the neurosurgeon. Nonetheless, surgery is the mainstay of glioma therapy because it provides the means of obtaining absolute tissue diagnosis, mapping tumor margins, mechanical cytoreduction, and guidance of adjuvant therapy such as brachytherapy, gene therapy, or radiosurgery. Recent developments in computation tec...
متن کاملProton Beam Therapy for Pediatric Brain Tumor
Cancer is a major cause of childhood death, with central nervous system (CNS) neoplasms being the second most common pediatric malignancy, following hematological cancer. Treatment of pediatric CNS malignancies requires multimodal treatment using a combination of surgery, chemotherapy, and radiotherapy, and advances in these treatments have given favorable results and longer survival. However, ...
